Beckman Coulter Cytoflex LX (21-color) Flow cytometry analyzer

The core facility offers a CytoFLEX LX flowcytometer which is a revolutionary system presenting optimal excitation and emission, minimizing light loss and maximizing sensitivity. The fully activated instrument includes three fluorescent channels from the 488 nm (Blue) laser, three from the  638 nm (Red) laser, five from the 405 nm (Violet) laser, five from the 561 nm (Yellow Green) laser, three from the  355 nm (UV) laser, and two from the 808 nm (Infrared) laser. 

The following colors can be measured:

1. 488 nm excitation; FITC, PerCP, PerCP-Cy5.5
2. 561 nm excitation; PE, ECD/mCherry, PECy5, PECy5.5, PECy7
3. 405 nm excitation; BV421, BV510, BV605, BV711 en BV786
4. 355nm excitation; BUV395, BUV496, BUV737
5. 638nm excitation; APC, Alexa700, APC-Cy7
6. 808nm excitation; AF790, PF840

Acquisition is possible with tubes and 96 (deep) well plates. Volumetric sample injection allows absolute cell counting. The system operates with the flexible Cytexpert software.

About our equipment

The technology center Flow cytometry holds nine flow cytometers (ranging from 4-colors up to 10-colors) and a flow sorter (18-color) that are available for research purposes. These are located within a ML-I/II biohazard room allowing analysis and sorting of genetically modified cells. A dedicated room is available for sorting viral-transduced and transfected cells with the required ML-I/II biosafety permit.

The research flow cytometers are supervised by senior specialists who train, advise and supervise internal and external personnel within the facility.

Using our equipment

Users who have operated flow cytometry equipment will, after evidence of proficiency, be allowed to use the equipment unassisted. Individual and group training in the use of our equipment is available to allow for unassisted use in the future.

    Reservations for sorting

    You can make a reservation request using your own Outlook agenda and invite the cell sorter.
    • In outlook select calendar (agenda) and then new (nieuw)
    • Select guide card invite attendees (deelnemers uitnodigen)  and send to (aan) Vz apparatuur Laboratoriumgeneeskunde LH flowcytometer Aria (VZ00329) and add this to required (vereist).
    • You can make this appointment. Select scheduling (planning). The bars indicate when the cell sorter is busy. Select the time you need to sort you samples.
    • Select appointment (afspraak). Mention your name and phone number under subject (onderwerp).
    • Finally, press send (verzenden).

Beckman Coulter Gallios Flow cytometry analyzer

The 10 color Beckman Coulter Gallios is a research system that delivers analytical excellence by coupling extraordinary sensitivity, resolution and dynamic range with high-speed datacollection.The system has 3 solid state lasers:
  1. 488nm excitation; FITC, PE, ECD, PC5, PC7
  2. 405nm excitation; Pacific Blue/BV421 and Krome Orange/BV510
  3. 642nm excitation; APC, APC-A700 and APC-A750
A selection from up to 62 parameters can be processed per analysis, at acquisition rates of 25,000 events-per-second. Acquistion software is Kaluza G (2014) and it functions with a carousel (32 tubes).

Sorters



Becton Dickinson Aria Flow cytometry sorter

The high-speed sorting cytometer is equipped with 5 lasers, allowing excitation with UV (355nm), 405,  488, 561, 640 nm. There are 18 fluorescence detectors, allowing analysis of 18 colors and FSC and SSC. This provides a powerful and flexible analysis platform suitable for a wide range of research applications. Cell subsets identified using this powerful analysis capability can if required be sorted, using an electrostatic deflection mechanism.

Analysis/sort rates of over 15000 cells per second are possible, and sorted cells (4 populations at the same time) may be collected in tubes, onto slides, or into the wells of microplates. There is a sample collection temperature control option available.The instrument is fitted with a robotic arm allowing the programmed deposition of specified numbers of particular cell types, for example into each well of a 96 well plate. Sorting purity should be >99%.
