In a recent publication in Scientific Reports, Wouter Verdurmen, Department of Biochemistry, Theme Nanomedicine and collaborators from the University of Zurich have utilized their newly developed biotin ligase assay for comparing the cytosolic uptake of a variety of protein transporters based on cell-penetrating peptides, bacterial toxins and supercharged proteins.

Effcient drug delivery is a major bottleneck for the development of novel therapies. Especially the ability of therapeutic agents to access the cytosol is of great importance for expanding the range of druggable targets. Even though several drug delivery systems have been reported to achieve cytosolic delivery, a side-by-side comparison using a quantitative method as the one published here, had never been done. The results underscore the effectiveness of bacterial toxin-based delivery systems.
