During fibrosis, local fibroblasts are activated to produce and deposit excess collagen and other extracellular matrix (ECM) proteins. Macrophages have been shown to respond to changes in their tissue microenvironment, however the molecular mechanisms underlying pro-fibrotic activation in response to these changes is yet to be elucidated. Here, we used in vitro 3D ECM hydrogels to mimic the fibrotic environment, and observed a macrophage phenotype akin to that seen in fibrotic mice. In our model, we uncovered a unique ECMmetabolic axis in macrophages, highlighting potential therapeutic targets for the treatment of fibrotic diseases.
